ondays1and8,balb/cmicereceived0.3%toluenediisocyanate(tdi)orthevehicleacetone–oliveoil(aoo)onthedorsumofbothears(20ml).onday14,themicewereoropharyngeallydosedwith40mlofanpsuspension(0.4mg?ml-1(,0.8mg?kg-1)tio2orau).1daylater(day15),themicereceivedanoropharyngealchallengewith0.01%tdi(20ml).onday16,airwayhyperreactivity(ahr),bronchoalveolarlavage(bal)cellandcytokineanalysis,lunghistology,andtotalserumimmunoglobulinewereassessed.
npexposureinsensitisedmiceledtoatwo-(tio2)orthree-fold(au)increaseinahr,andathree-(tio2)orfive-fold(au)increaseinbaltotalcellcounts,mainlycomprisingneutrophilsandmacrophages.thenpstakenupbybalmacrophageswereidentifiedbyenergydispersivex-rayspectroscopy.histologicalanalysisrevealedincreasedoedema,epithelialdamageandinflammation.
inconclusion,theseresultsshowthatalow,intrapulmonarydosesoftio2oraunpscanaggravatepulmonaryinflammationandahrinamousemodelofdiisocyanate-inducedasthma.
